  为搭建我校博士后之间的学术交流平台,促进学术水平提升,学校博士后管理办公室组织开展博士后学术沙龙活动。本次沙龙由我校博士后梁鸿、胡兰琳和杨业明分享其研究成果,诚挚邀请感兴趣的师生参加。

  报告一:     

  (1)主 题:Design of novel supramolecular self-assembly gene vector

  (2)主讲人:梁鸿  电子科技大学医学院博士后

  (3)交流内容:

  The global scale of gene therapy is growing rapidly at a compound annual growth rate of about 65%. It will grow from $1 billion in 2017 to $44 billion in 2024. According to the clinical trials of gene therapy in the world, tumor treatment accounts for 65% of all disease treatments. Dr. Liang focused on constructing non-viral gene vector for tumor theranostics. Based on supramolecular theory, he has designed a series of amphiphilic dendritic lipid polypeptide molecules that could be self-assembly to form nanoparticles. These nanoparticles have excellent gene transfection effect that was 58.7% greater than PEI in the condition of 10% fetal bovine serum for HeLa cells. And He also achieved accurate imaging-guidance tumor therapy with intelligent “turn-on” fluorescence imaging in vivo. In this salon, He will share the design ideas and experimental data of these novel supramolecular self-assembly nanoplatform.

  (4)主讲人简介:

  Dr. Liang received his doctor's degree of Biomedical Engineering from National Engineering Research Center for Biomaterials of Sichuan University in 2019. His main research field is self-assembly nanoplatform for tumor theranostics. At present, He has published 8 SCI papers including 5 papers as the first author and the total impact factor exceeds 50. These papers mainly published in internationally renowned journals in the relevant field such as Small, Chemical Communications, ACS Applied Materials & Interfaces, Journal of Chemistry Materials B, Chinese Journal of Chemistry. He has already owned obtained 3 authorized Chinese invention patent. He used to work in the medical information center of the KeLun Institute of Medicine. In 2021, he became a full-time postdoctoral fellow jointly trained by UESTC and Sichuan Provincial People's Hospital.

  (1)主 题:Molecular Mechanisms of Metastasis in NSCLC

  (2)主讲人:胡兰琳 医学院博士后

  (3)交流内容:

  Lung cancer is a common malignancy with high morbidity and mortality. The traditional therapeutic managements of non-small cell lung cancer (NSCLC) include surgical resection, radiotherapy, chemotherapy, and targeted therapy. In the past decade, the emergence of immunotherapy, especially immune checkpoint inhibitors (ICIs), has greatly changed the therapeutical landscape of NSCLC. Nevertheless, the metastasis and recurrence remain main causes of failure in NSCLC treatment. To investigate the molecular mechanisms of metastasis in lung cancer, we utilized a trans-well invasion system introduced by Chu et al. to conditionally select the minor population of cells with high aggressiveness (Chu et al., 1997). RNA-seq was then performed to analyze the differentially expressing genes and potential signaling pathways in this group of cells. We found several genes play roles in inducing invasion in NSCLC cell lines. More detailed mechanisms are under investigation currently.

  (4)主讲人简介:

  Lanlin Hu received her B.S. in Biological Engineer and Ph.D. in Cell Biology from Sichuan University. During Ph.D. program, she received the scholarship from the China Scholarship Council and has studied for two years in Head and Neck Lab in University of California at San Francisco. She is currently a post-doc advised by Prof. Chuan Xu in School of Medicine of UESTC, with the interests in understanding the molecular mechanisms of heterogenicity in cancer.  

  报告三:

  (1)主 题:A missense mutation in Pitx2 leads to early-onset glaucoma via NRF2-YAP1 axis

  (3)交流内容:

  Glaucoma, affecting 70 million people worldwide, is a group of neurodegenerative diseases characterized by the progressive death of retinal ganglion cells (RGCs) and atrophic excavation of the optic nerve, ultimately resulting in irreversible loss of vision. A genome-wide scan of pooled DNA from 12 affected and 12 unaffected mice was carried out using 48 microsatellite markers. The causative mutation was identified by comparing the whole exome sequences from a homozygous mutant and control. We then generated a Pitx2 knock-in mutant mouse model by introducing the point mutation c. 344G>T in exon 2.

  The mutation causes an elevation in IOP and progressive death of retinal ganglion cells (RGC). These ocular phenotypes recapitulate features of pathologies observed in human glaucoma. Increased oxidative stress was evident in the inner retina. We demonstrate that the mutant PITX2 protein was not capable of binding to Nuclear factor-like 2 (NRF2), which regulates Pitx2 expression and nuclear localization, and to YAP1, which is necessary for co-initiation of transcription of downstream targets. PITX2-mediated transcription of several antioxidant genes were also impaired. Treatment with N-Acetyl-L-cysteine exerted a profound neuroprotective effect on glaucoma associated neuropathies, presumably through inhibition of oxidative stress. Our study demonstrates an essential role of PITX2 in glaucoma optic pathogenesis. We also provide a novel early-onset glaucoma model and a corresponding therapeutic strategy.

  (4)主讲人简介:

  Yeming Yang received the bachelor’s degree from Qingdao University and Ph.D. degree from the UESTC of Biomedical engineering, in 2015 and 2020, respectively. He currently works as a post-doc advised by Prof. Xianjun Zhu in the of Medical School of UESTC. He is working in the field of function study of disease genes. His main research interests include neuroscience, mouse genetics and oculopathy research.
